Background: Mood dysregulation disorders and insomnia have become a global public health crisis. Conventional pharmacological approaches often face limitations in the form of adverse side effects and dependency risks. The Endocannabinoid System (ECS), particularly through the endogenous ligand Anandamide (AEA), offers a significant alternative therapeutic target. However, AEA's half-life is very short due to degradation by the Fatty Acid Amide Hydrolase (FAAH) enzyme. Indirect modulation using botanical agents has the potential to overcome this issue more safely. Methods: This study collects and synthesizes clinical and preclinical methodologies from existing literature. Evaluated parameters include in vivo microdialysis for sleep architecture, animal neurobehavioral testing (such as the Elevated Plus Maze, Open Field Test, Forced Swimming Test, and Pentobarbitone Sleep Potentiation), as well as randomized controlled trials (RCTs) in humans measuring cortisol biomarkers, Heart Rate Variability (HRV), and psychometric mood evaluations. Results: Phytochemical compounds in cacao, specifically N-oleoylethanolamine (OEA) and N-linoleoylethanolamine (LEA), act as competitive FAAH inhibitors. On the other hand, nutmeg isolates such as 5'-methoxylicarin A, Licarin A, and Malabaricone C demonstrate highly potent FAAH inhibition at the micromolar level. Both significantly prolong AEA circulation. Preclinically and clinically, these interventions are proven to trigger adenosine increases that extend Slow-Wave Sleep (SWS), reduce cortisol excretion, and decrease hyperarousal responses to autonomic stress. The combination of both creates a dual-pathway allosteric blockade on FAAH. Conclusion: The combination of Cacao (Theobroma cacao) and Nutmeg (Myristica fragrans) extracts represents a promising non-pharmacological biotherapy breakthrough. The pharmacokinetic synergism and entourage effect of both works effectively in managing the endogenous anandamide circuit to stabilize the neuro-emotional axis and improve sleep quality without posing long-term toxicity risks.
